ThiskitenablesclusteramplificationonthecBotSystem.DNAlibrarysamplesareboundtocomplementaryadapteroligosgraftedonthesurfaceoftheflowcell.Thetemplatesarecopiedfromthehybridizedprimerby3"extensionusingahighfidelityDNApolymerase.Thesecopiesareisothermallyamplifiedtocreateclonalclustersof~1,000copieseach,readyforsequencing.
Clustergenerationreagentsareprovidedinapre-mixed,96-wellplateformatthatrequiresminimalreagentpreparation.Thekitalsoincludesreagentsforclusterresynthesisofthereversestrand,regeneratedbybridgeamplificationwithinthepaired-endflowcell.Afterresynthesisofthereversestrand,theoriginalforwardstrandiscleavedandthereversestrandissequencedforthesecondread.
